چکیده

Cytosol trafficking is a limiting step of viral infection or DNA delivery. Starting from the cell surface, most viruses have to travel through a crowded and risky environment in order to reach a small nuclear pore. This work is dedicated to estimating the probability pN of a viral arrival success and, in that case, the mean time τN it takes. Viral movement is described by a stochastic equation, containing both a drift and a Brownian component. The drift part represents the movement along microtubules, while the Brownian component corresponds to the free diffusion. The success of a viral infection is limited by a killing activity occurring inside the cytoplasm. We model the killing activity by a steady state killing rate k. Because nuclear pores occupy a small fraction of the nuclear area, we use this property to obtain asymptotic estimates of pN and τN as a function of the diffusion constant D, the amplitude of the drift B and the killing rate k.

Intercellular Trafficking of VP22, a Herpes Simplex Virus Type 1 Tegument Protein

The herpes simplex virus type 1 (HSV-1) tegument protein, VP22 has been reported to have the property of intercellular transport. The previous studies have shown that following expression of a fusion protein containing VP22 it spreads to every cell in a monolayer and concentrates in the nucleus. In spite of these reports, some studies have shown that VP22 trafficking and its nucleus accumulatio...

Modeling the Early Steps of Cytoplasmic Trafficking in Viral Infection and Gene Delivery

Gene delivery of nucleic acid to the cell nucleus is a fundamental step in gene therapy. In this review of modeling drug and gene delivery, we focus on the particular stage of plasmid DNA or virus cytoplasmic trafficking. A challenging problem is to quantify the success of this limiting stage. We present some models and simulations of plasmid trafficking and of the limiting phase of DNA-polycat...

Bovine Leukemia Virus Presence in Breast Tissue of Argentinian Females and Its Association With Cell Proliferation and Prognosis Markers

Abstract Introduction: Bovine leukemia virus (BLV) causes enzootic bovine leukemia, andis closely related to human T-lymphotropic virus type 1. It expresses microRNAs of unknown function and codes Tax, the protein that mediates malignant transformation. BLV is capable of infecting B- and T-lymphocytes, endothelial cells, and mammary epithelial cells of cattle. Several studies demonstrated the ...
